چکیده

The plaque assay has long served as the "gold standard" to measure virus infectivity and test antiviral drugs, but the assay is labor-intensive, lacks sensitivity, uses excessive reagents, and is hard to automate. Recent modification of the assay to exploit flow-enhanced virus spread with quantitative imaging has increased its sensitivity. Here we performed flow-enhanced infection assays in microscale channels, employing passive fluid pumping to inoculate cell monolayers with virus and drive infection spread. Our test of an antiviral drug (5-fluorouracil) against vesicular stomatitis virus infections of BHK cell monolayers yielded a two-fold improvement in sensitivity, relative to the standard assay based on plaque counting. The reduction in scale, simplified fluid handling, image-based quantification, and higher assay sensitivity will enable infection measurements for high-throughput drug screening, sero-conversion testing, and patient-specific diagnosis of viral infections.

Replication Kinetic of Infectious Laryngotracheitis Virus in Embryonic Chicken Neural Stem Cell

Background and Aims: Infectious laryngotracheitis virus (ILTV) infection causes a highly contagious respiratory disease in poultry leads to significant economic losses in the poultry industry. The genomic and antigenic characteristics between virulent and vaccine strains of ILTV are very similar. The ability of embryonated chicken neural stem (CNS) cell for differential diagnosis of ILTV strain...

Interfering With Lipid Raft Association: A Mechanism to Control Influenza Virus Infection By Sambucus Nigra

Sambucus nigra (elder) are broadly used species to treat microbial infections. Thepotential antiviral activity and mechanism action of elder fruit (EF) in human epithelium cell(A549) cultures infected with H9N2 influenza virus were determined. The effect of variousconcentrations of EF on influenza virus replication was examined by using virus titration, quantitative real time RT-PCR, fusion and ...

Using Boehmite Nanoparticles as an Undercoat, and Riboflavin as a Redox Probe for Immunosensor Designing: Ultrasensitive Detection of Hepatitis C Virus Core Antigen

In this study a label-free electrochemical Immunosensor for ultrasensitive detection of Hepatitis C virus core antigen in serum samples was fabricated by using a simple approach. In this method a low-cost and sensitive immunosensor was fabricated based on a boehmite nanoparticles (BNPs) modified glassy carbon. The BNPs provide a specific platform with increased surface area which is capable of ...

Comparison of MTT and Trypan Blue methods in determining the survival of Vero cell line in HSV-1 infection

Abstract Background and Objectives Evaluation of cell survival is important in cellular studies and the result is decisive for the application of the test substance. Different methods are used to evaluate cell survival and the correlation between methods is important. Here, Vero cell line was used to evaluate the effects of HSV-1 virus on cell survival. Cell survival after HSV-1 infection usin...
